An analytic model compared SARS-CoV-2 screening strategies over an 80-day abbreviated college semester for a hypothetical cohort of 4,990 uninfected students and 10 students with undetected asymptomatic infections. It varied screening frequency, test sensitivity and specificity, and costs from \$10 to \$50 per test. The model identified screening every two days, combined with strict behavioral interventions, as the preferred strategy for balancing outbreak control and cost, even when rapid tests had low sensitivity, provided specificity was high.
